A recently granted patent (Publication Number: US11859254B2) discloses a method for measuring the methylation level of a CpG site for three or more markers in a biological sample of a human individual. The method involves treating genomic DNA in the sample with bisulfite, amplifying the bisulfite-treated genomic DNA using specific primers for each marker, and determining the methylation level through various techniques such as methylation-specific PCR, quantitative methylation-specific PCR, and bisulfite genomic sequencing PCR. The markers used in the method include ELMO1, ZNF569, C13orf18, CD1D, ARHGEF4, SFMBT2, PPP2R5C, CYP26C1, PKIA, CLEC11A, LRRC4, ST8SIA1, NDRG4, and FER1L4, with SFMBT2, FER1L4, and ARHGEF4 being mandatory markers.

Furthermore, the patent details specific primer sequences for each marker, enabling the binding of amplicons for accurate measurement of methylation levels. The biological sample can be derived from various sources such as tissue, stool, urine, blood, plasma, or serum. Additionally, the CpG site targeted for methylation analysis can be located in a coding or regulatory region. The method allows for the determination of methylation scores or frequencies for the CpG sites, providing valuable insights into epigenetic modifications in the individual’s genome. This innovative approach offers a comprehensive and precise method for assessing methylation levels in multiple markers simultaneously, enhancing the understanding of epigenetic mechanisms in human biology.
